Abstract
This article is devoted to the development of a measurement procedure of the cell number concentration in liquids. This procedure is intended to determine the number of viable microorganisms, such as Bifidobacterium, Lactobacillus acidophilus, Listeria monocytogenes, Cronobacter sakazakii, Clostridium perfringens, Staphylococcus aureus, Proteus, Enterococcus, coliforms, Vibrio parahaemolyticus, and Bacillus cereus, in liquids. This methodology implements two main measurement methods: the Petri dish method and direct counting using an inverted fluorescence microscope. It describes the conditions and procedures that ensure measurement accuracy, the scope of the methodology, and the new approaches that enabled the development of a methodology with the best measurement accuracy available for these methods.
The established measurement range for cell number concentration varies from 1 to 1 · 105 1/cm3, allowing the methodology to be applied to a variety of samples, including both highly diluted and concentrated solutions.
The methodology’s application areas include healthcare, veterinary medicine, and the food industry, making it an important tool for quality control of laboratory analyses and food safety assessments. The developed methodology can serve as the basis for the creation of reference materials for cell number concentration, which will facilitate the standardization of measurement processes and increase confidence in the analysis results among consumers and regulatory authorities.
This article makes a significant contribution to the development of measurement methods in microbiology and can be useful for both scientific research and practical applications in various fields.
